Rotherham endured a dismal 2023-24 season, finishing rock bottom of the Championship as they confirmed their relegation back to League One.
The Millers would have been hoping for a more successful return to the third tier, currently placed 14th in the standings and unlikely to reach the playoff places this term.
Steve Evans's men have won 11, drawn eight and lost 14 of their 33 league matches, leaving them with 41 points - 15 away from sixth-placed Charlton Athletic and only eight above the drop zone.
They did recently end a miserable run of five losses and one draw by defeating Bristol Rovers 3-2 in their last match, a result they will be aiming to build on here.
While Rotherham are hosting a Leyton Orient side that are challenging for the playoff places, the U's have faltered in the past weeks.
Leyton Orient secured three consecutive victories, including a superb win against Wrexham, before falling to three successive defeats leading into this one.
Those results have seen them fall out of the playoff places and down to eighth, though they could move level on points with sixth-placed Charlton if they win here and Charlton lose.
The visitors have already defeated Rotherham this season, securing a 1-0 win in the reverse fixture, and they will look to replicate that result on Tuesday.

The hosts could be without the services of Mallik Wilks after the attacking midfielder was substituted due to injury in the last outing, meaning Jack Holmes may start here.
Elsewhere, the remainder of the side that twice came from behind to eventually defeat Bristol Rovers could start on Tuesday, including top scorer Sam Nombe, who scored twice in that game.
As for Leyton Orient, a devastating defeat to Charlton saw them concede two stoppage time goals, and Richie Wellens may make several changes as a result.
